B12 is an east-west route through eastern South Australia, linking Adelaide (via Princes Highway) with a number of small Mallee towns and north-western Victoria. Since the opening of the Adelaide-Crafers Highway project in 2000, the route has grown in importance and popularity as a shorter route between Sydney and Adelaide, bypassing the Riverland towns.

The route is constructed generally as a high-standard rural two-lane highway. It passes through some sparsely populated areas, although there are a number of small hamlets dotted along the railway line which parallels the Mallee Highway.

Prior to 1998/99, the Mallee Highway was signposted as National Route 12 which extended into Victoria as far as Ouyen and, later, Tooleybuc. As part of the introduction of alpha-numeric route marking in South Australia, B12 replaced National Route 12.
